Effects of Ginseng Butanol Fraction on the Development of Morphine Tolerance and Physical Dependence in Mice





Abstract
To evaluate the effects of Ginseng Butanol Fraction(GBF) on morphine tolerance and physical dependence in mice this experiment was carried out. Experimental groups were divided into Control, GBF pretreatment and Naloxone group. Control groups were subdivided into saline group which was injected 0.9% saline 3 times daily, GBF Control group which was injected 50, 100 and 200mg/kg once daily for 4 days, respectively. And Morphine group which was injected morphine HC1 3 times daily by 50, 50, and 75mg/kg with daily increase by 25mg/kg in each injection. GBF pretreatment group was injected GBF by 50, 100 and 200mg/kg once daily for 4 days, respectively, then followed by scheduled morphine injection. To evaluate the effects of GBF pretreatment on the body temperature, analgesia and weight in chronic morphinized mice, the experimental observations were done after additional morphine by 5mg/kg after last morphine injection, respectively. In Naloxone group administered by 1mg/kg in the group of morphinized, GBF pretreatment groups were observed for the frequency of jumping at 5hr after daily last injection of morphine. All experimental observations were done under the 15¡¾1¡ÆC ambient temperature.
The results obtained were as follows
1. Morphine-induced hypothermia was inhibited by a pretreatment with GBF 200mg/kg
2. Inhibition of morphine tolerance by a pretreatment with GBF 200mg/kg was proved by enhancement of analgesic effect after additional morphine injection.
3. Pretreatment of both GBF 50mg/kg and 200mg/kg reduced the loss of body weight occurring when the morphine administration was terminated.
4. The characteristic jumping response, the abstinence sign manifested by Naloxone injection in morphinized mice were promoted by a pretreatment with GBF.
Therefore, the pretreatment of GBF inhibited the development of tolerance and inhibited partially the development of physical dependence in morphinized mice. But Naloxone induced jumping effects was not inhibited by GBF.
